Histone Deacetylases and Their Potential as Targets to Enhance Tumour Radiosensitisation

چکیده

In mammalian cells, genomic DNA is packaged with histone proteins and condensed into chromatin. To gain access to the DNA, chromatin remodelling required that enhanced through post-translational modifications, which subsequently stimulate processes including repair transcription. Histone acetylation one of most well understood modifications controlled by acetyltransferases (HATs) deacetylases (HDACs). These enzymes play critical roles in normal cellular functioning, dysregulation HDAC expression particular has been linked development a number different cancer types. Conversely, tumour cell killing following radiotherapy triggered damage HDACs can help co-ordinate response promotes radioresistance. Consequently, inhibitors have investigated as potential radiosensitizers vitro vivo improve efficacy or specific this review, we provide an up-to-date summary their functions, repair. We also review evidence demonstrating effectively enhance radiosensitisation, therefore show for translation clinic patient benefit.

treatment regimen is still an unmet need. This might lead to the fact that few CCA patients are suitable for the surgery as well as the limitation of other therapeutic modalities (Patel, 2006; Sia et al., 2013; Zabron et al., 2013). In addition, the palliative chemotherapy and radiotherapy have been used as adjuvant therapy for this cancer type. Various chemotherapeutic agents e.g. 5-fluorourac...

DNA and histone deacetylases as targets for neuroblastoma treatment

Neuroblastoma, a tumor of the peripheral sympathetic nervous system, is the most frequent solid extra cranial tumor in children and is a major cause of death from neoplasia in infancy. Still little improvement in therapeutic options has been made, requiring a need for the development of new therapies. In our laboratory, we address still unsettled questions, which of mechanisms of action of DNA-...

Histone deacetylases as targets for treatment of multiple diseases.

HDACs (histone deacetylases) are a group of enzymes that deacetylate histones as well as non-histone proteins. They are known as modulators of gene transcription and are associated with proliferation and differentiation of a variety of cell types and the pathogenesis of some diseases. Recently, HDACs have come to be considered crucial targets in various diseases, including cancer, interstitial ...

Integrins as Determinants of Genetic Susceptibility, Tumour Behaviour and Their Potential as Therapeutic Targets

Metastatic prostate tumours are responsible for the majority of deaths associated with prostate cancer. The most frequent site of prostate cancer metastasis is to bone; over 80% of men who die of the disease have metastatic bony lesions (Bubendorf et al., 2000). Early detection of prostate cancer remains crucial to effective treatment. However, we are still unable to identify with certainty tho...
